At the EHF Cup 2000/01 57 took Handball part -Vereinsmannschaften, who had qualified in the previous season in their home leagues for the competition. It was the 20th edition of the EHF Cup or the IHF Cup. The cup games began on August 30, 2000, the second final took place on April 28, 2001. In the final, SC Magdeburg was able to prevail against the defending champion RK Metković .
All rounds including the final were carried out in a knockout system with a return leg. In round 1, 18 lower-rated clubs started the competition. In round 2 there were another 15 teams who had also qualified in their home leagues in the previous season, and the 8 losers in qualifying round 1 from the EHF Champions League 2000/01 . In round 3, a further 8 higher-rated teams including the defending champion joined the team, plus the 8 losers from qualifying round 2 from the Champions League.
