The order for the boat was awarded to the Vegesacker shipyard in Bremen on August 7, 1939 . The keel was laid on August 21, 1940, the launch on April 28, 1941. The commissioning under Lieutenant Hermann Hesse finally took place on July 5, 1941.

The boat went into service from July 5 to September 30, 1941 as a training boat for the 7th U-Flotilla in Kiel . After training, U 133 belonged to the 7th U-Flotilla in St. Nazaire as a front boat from October 1, 1941 to December 31, 1941 , and from January 1, 1942 to its sinking on March 14, 1942 as a front boat to the 23 U-flotilla in Salamis .

The wreck was rediscovered by Greek professional divers Efstáthios Baramátis and Theófilos Klímis at a depth of 74 meters in 1986. The submarine is still in the marine grid square CK 8948.
