Minamisaku-gun was created on January 14, 1879 by dividing the old Saku-gun into Minamisaku-gun and Kitasaku-gun ("North Saku-gun"). The seat of the then district administration was Usuda. As of April 1, 1889, 23 villages belonged to Minamisaku-gun. On June 30, 1893 Usuda and on March 9, 1897 Nozawa was appointed city. On November 1, 1919, Nakagomi was appointed city. Until the major territorial reform of the 1950s, three cities and 16 villages belonged to Minamisaku-gun. This major territorial reform reduced the number of municipalities belonging to Minamisaku-gun to three cities and five villages by April 1, 1961. Two further amalgamations on March 20 and April 1, 2005 reduced the number to the current one.
