Pereyra began his career in 2009 at CA River Plate , where he played for three years in his youth. After he came to use in the second half of his first professional season, he received regular playing times in the following season. In his second full season for River Plate, Pereyra was a regular and played 29 games. In 2011 he was signed by the Italian first division club Udinese Calcio . In his first season in Italy, in which he only played eleven games, he was able to reach third place with Udinese and thus qualify for the Champions League , in which however they were defeated by Sporting Braga . In the 2012/13 season , Udinese, this time with Pereyra as a regular player, qualified for the international competition again in fifth place.

After a disappointing season , at the end of which they finished 13th and long-time coach Francesco Guidolin resigned, Pereyra's next career step was after 84 league games and eight goals for Udinese: for a fee of 1.5 million euros, he switched to a loan with an option to buy to Juventus Turin . With Juve, Pereyra won the championship and the cup under Massimiliano Allegri and was in the final of the Champions League . He came to 35 league (four goals), four cup (two goals) and twelve Champions League appearances (no goal). For the 2015/16 season , Pereyra was finally committed for 14 million euros and given a contract until June 30, 2020.

In August 2016 Roberto Pereyra moved to Watford FC in the English Premier League , where he received a five-year contract. The transfer fee was 13 million euros plus possible bonuses of two million euros.
