Within St. Louis County, Jefferson Township borders on Gravois Township in the south, Bonhomme Township in the west, Clayton Township in the north and northwest and Hadley Township in the northeast .

The Interstate 44 , St. Louis, Oklahoma City connects, runs through the center of Jefferson Township. Missouri State Route 100 runs through the north of the township ; the Missouri State Route 366 , on a part of the track of the old route 66 extends, forms the southern boundary of the Township. All other streets within the township are inner-city connecting streets.

In Jefferson Township, the western terminus of the lies Blue Line of the Metrolink called Light rail -Nahverkehrssystems the metropolitan area around St. Louis.

A railway line of the BNSF Railway runs from St. Louis in a south-westerly direction through the Jefferson Township , via which Amtrak passenger trains also run.

The Lambert-Saint Louis International Airport is located about 20 km north of Jefferson Township.
