In the Bantam Draft of the Western Hockey League (WHL), one of the three highest junior leagues in Canada, he was then selected in 2007 as the first player in the history of the newly formed Edmonton Oil Kings in third overall position. In his second full season he was appointed assistant captain of the Oil Kings in 2009/10 and ended the season with 24 points from 48 games. Although he missed almost a third of the season with a foot injury, he was voted Most Valuable Player on his team. In the 2010 NHL Entry Draft , the Buffalo Sabers then selected him in the first round in 23rd position.
In the following two seasons Pysyk acted as captain of the Oil Kings and led them to win the title in the WHL in the 2011/12 season . With eight assists, he was the best assists among the defenders of his team in the play-offs .
For the 2012/13 season Mark Pysyk made his professional debut for the farm team of the Buffalo Sabers, the Rochester Americans , in the American Hockey League . In his rookie season, he was called up for the first time in the NHL squad of the Sabers and made his debut in March 2013 in the game against the Washington Capitals . By the end of the season, Pysyk played 19 games and scored five points scorer. He switched frequently between Buffalo and Rochester in the following two seasons before signing a new two-year deal with the Sabers in July 2015. At the beginning of the 2015/16 season he established himself alongside Jake McCabe . Both were praised for their high mobility and their overview of the game.
In June 2016, Pysyk was given two voting rights for the NHL Entry Draft 2016 to the Florida Panthers , who in return transferred Dmitri Kulikow to Buffalo.
