In Montana, Duke began playing poker at card shops in Billings . Her brother Howard Lederer was already a professional player at this point and supported his sister financially. In 1994 she moved to Las Vegas with her husband so that she could devote herself to poker. Prior to 2004, she reached the 10th place in the Main Event of the World Series of Poker (WSOP) in 2000 , while she was in the eighth month of pregnancy with her third child. In early 2004, Duke gained some notoriety for coaching actor Ben Affleck , who subsequently won the California State Poker Championship . In September 2004, Duke won the WSOP Tournament of Champions for a $ 2 million prize money . She prevailed as the only woman at the final table against the then world champion Greg Raymer and in heads-up against Phil Hellmuth . After this success, she appeared on the Late Show with David Letterman . In the 2004 WSOP won Duke a bracelet in the variant Omaha Hi-Lo .

Duke no longer takes part in women’s tournaments, as she says poker is one of the few sports where women have exactly the same chances as men, making special women’s tournaments unnecessary. Duke is one of the many poker players who doesn't like the restrictions on television tournaments. Although players have to pay an entry fee, many organizers do not allow players to wear sponsor logos. In April 2005, Duke sparked controversy when she remarked that poker players were not even slaves, but had to pay to work on plantations (original: We [poker players] are not even slaves. We're people paying to pick the cotton. ).

Overall, Duke has earned more than $ 4 million in live poker tournaments, making it fourth among the most successful women in terms of tournament prize money .

She appeared as the first athlete on Comedy Central's The Colbert Report on January 30, 2006 , where she also discussed her book and what it's like to assert yourself as a woman in a male-dominated sport. At the end of the show, presenter Stephen Colbert celebrated a victory over Duke in a game of five-card draw before realizing that a new deck of cards had never been shuffled.

In 2002 she moved to Portland , Oregon to work for ieLogic. This company produces software for online poker. In 2004 she divorced her husband, Ben Duke.
