Fernando Muslera was born as the son of the waiter Hugo and the housewife Norma Muslera. Together with his parents and his sister Carolina, he grew up in the Barrio Lavalleja in Montevidean until he was 16 , after which the family moved to the Barrio Prado . Fernando started playing football at Club Ferrocarril Oeste at the age of four , later moving to Royal before eventually joining Cosmos . He played as a center forward until he was eight. At the age of twelve he switched to the Wanderers , where he also began his professional career. On January 12, 2007 he moved to Nacional Montevideo on loan for a year . A rental fee was not due. In the summer of 2007, several top European clubs became aware of the talented goalkeeper. The Italian Champions League participant Lazio Rome finally secured his services for three million euros. Muslera came on as a replacement for Angelo Peruzzi , who had ended his career in the summer.

He made his Serie A debut on September 16, 2007 in a 2-2 draw against Cagliari Calcio . In his first season in Italy he was only a substitute for the oldest player in the league, Marco Ballotta, after mistakes in the game against Inter Milan (1: 5) . After his contract ended in summer 2008, he was able to prevail as a regular goalkeeper.

On July 19, 2011 it was officially announced that Muslera was moving to Turkey to Galatasaray Istanbul . He signed a five-year contract there. On April 8, 2012, the last matchday of the season, he scored his first league game goal for Galatasaray with a penalty in the 83rd minute against Manisaspor .

Muslera won 13 national trophies with Galatasaray Istanbul until the 2018/19 season and was able to establish itself as a fan favorite. He is also the foreigner with the most appearances for Galatasaray. On matchday 27 of the 2019/20 season, Muslera was seriously injured in a collision with opponent Milan Škoda from Çaykur Rizespor . The goalkeeper sustained a broken tibia and fibula.
