In the final, three Americans met two Jamaicans and one athlete each from the Ivory Coast, France and Trinidad and Tobago.
The favorite was the 2008 Olympic champion and reigning world champion Veronica Campbell-Brown from Jamaica. The strongest competitor was the US athlete Allyson Felix, silver medalist in 2008 and third in the last world championships. The Jamaican Shelly-Ann Fraser-Pryce, winner of the 100 meters , and the US runner Sanya Richards-Ross, Olympic champion over 400 meters , also had medal chances .
Felix got off to the best start and quickly equalized the cornering requirement on Murielle Ahouré from Ivory Coast. At the start of the home stretch she was in the lead, closely followed by her teammate Carmelita Jeter. Jeter could no longer follow her compatriot and was also overtaken by Fraser-Pryce. Felix won with 21 hundredths of a second ahead of Fraser-Pryce, who was 5 hundredths of a second ahead of Jeter. Campbell-Brown crossed the finish line in fourth, Richards-Ross fifth ahead of Ahouré.
With the two medals from Allyson Felix and Carmelita Jeter, the USA drew level with Jamaica in the overall statistics. Both teams had won eleven medals each in the previous seventeen Olympic finals. For the US team it was the sixth victory in this discipline.
