Preuilly-sur-Claise is located in the far west of the Touraine on the borders of Bas-Berry and Poitou . The districts are: La Berjaudière, Les Blanchards, La Chatrie, Les Chauvraux, Les Chirons, La Grange au Moine, Maupertuis, La Parentière, Le Pontreau, Pouplouroux, La Presle, Saint-Michel-du-Bois (incorporated in 1814) and La Touche south of the Claise and Champeaux, Les Effes, La Fichardière, La Folie, Fontbaudry, Les Martinières, Le Pouet, Les Ragots and Les Vigneaux north of the river. Neighboring municipalities of Preuilly-sur-Claise are: Boussay in the west, Le Petit-Pressigny in the north-west, Charnizay in the north, Bossay-sur-Claise in the east, Yzeures-sur-Creuse in the south, and Boussay in the south-west.
