Pepe began his career in his hometown of Maceió with SC Corinthians Alagoano . At the age of 18, he left Alagoas on his own and went to Portugal, where he played for various clubs before he was signed by the Portuguese first division club Marítimo Funchal . There he quickly conquered a regular position in central defense alongside Mitchell van der Gaag . In July 2002 he trained with Sporting Lisbon for two weeks , but a contract was not concluded for financial reasons.

After a three-year stay, he finally came to FC Porto in 2004 for around two million euros . With this Pepe celebrated numerous successes. In 2004 he won the World Cup , in 2006 he achieved the double of championship and cup victory as well as victory in the Supercup. In the 2006/07 season they won the championship again. He was naturalized in Portugal in 2007 and has played for the Portuguese national soccer team ever since .

In the summer of the same year he moved to Spanish record champions Real Madrid for a sum of around 30 million euros . In his first season he won the Spanish championship.

On April 21, 2009, Pepe lost control in the Madrid league game against Getafe FC after a foul on his opponent Casquero in the penalty area. In his opinion, the Getafe player had deliberately dropped. After the referee's whistle, he kicked the player who was lying on the ground several times, pushed him to the ground and hit Juan Albín in the face. Pepe was sent off with the red card and cursed the referees when leaving the field. After the game he was suspended for ten league games.

In the following years, Pepe was repeatedly noticed by excessive fouls and unsportsmanlike conduct. For example, in the first leg of the Spanish Cup quarter-finals in January 2012, he apparently deliberately stepped on the hand of Lionel Messi, who was lying on the ground . In the media he came under heavy criticism for his behavior several times. In total, Pepe was sent off six times for Real Madrid.

In the 2009/10 season , Pepe fell out due to injury and only played ten league games. After his recovery, he fought his way back into the team with his headball strength and established himself as a regular player and one of the world's best central defenders. In July 2011, he extended his expiring contract at Real Madrid for another five years until 2016. In the 2011/12 season , he made 29 appearances to win the championship. Two years later, he won the Champions League with Real Madrid and was selected by UEFA for his team of the season.

After two further Champions League titles (2016, 2017), another championship ( 2017 ) and two successes each in the UEFA Super Cup and the FIFA Club World Cup , Pepe announced at the end of the 2016/17 season after ten years of club membership and expiration his contract his departure from Real Madrid. The club management had only offered him a contract extension for another year. Pepe had refused this.

For the 2017/18 season , Pepe signed a two-year contract with Beşiktaş Istanbul . In mid-December 2018, after mutual agreement with the club's management, Pepe prematurely terminated his contract and left the club.

In early January 2019, Pepe returned to FC Porto . He received a contract with a term until June 30, 2021.
