All three Americans had qualified for the final. The final field was completed by one starter each from Barbados, Ghana, Jamaica, Portugal and St. Kitts and Nevis.

The Olympic champion from Sydney Maurice Greene from the United States met the reigning world champion Kim Collins from St. Kitts and Nevis and the reigning European champion , the Nigerian-born Francis Obikwelu, who started for Portugal. Jamaican Asafa Powell was one of the favorites.

The final race saw the closest decision in Olympic history to date. The runners all got off their starting blocks well and the field was extremely close. Nobody could break away and finally the US sprinter Justin Gatlin became Olympic champion in 9.85 s. He was a hundredth of a second ahead of Obikwelu, who was a hundredth of a second ahead of Greene. Another two hundredths of a second behind was the third US finalist Shawn Crawford and five hundredths of a second after Crawford the Jamaican Asafa Powell crossed the finish line in fifth. World champion Kim Collins was sixth with exactly 10.00 s.

In the 25th Olympic final, Justin Gatlin won the 16th gold medal for the USA in this discipline.

Francis Obikwelu won Portugal's first Olympic medal over 100 meters .
