Highway 24 begins in Colorado not far from the town of Minturn at the junction of Highway 6 with Interstate 70. The road leads south and crosses the continental divide with the 3117 m high Tennessee Pass . Further south, Highway 24 runs through Leadville and turns three miles south of Buena Vista along with US Highway 285 to the northeast.

After Highway 285 has left the common route to the north, Highway 24 reaches the city of Colorado Springs after several loops in northeast and southeast direction . There, Highway 24 meets Interstate 25 and US Highways 85 and 87 . After crossing the city from east to west, Highway 24 passes the airport and leaves the Colorado Springs metropolitan area in a northeastern direction.

In Limon , the road meets the US Highways 40 and 287 and again the Interstate 70, with which the Highway 24 leads partly on a common route and partly parallel to it to the east. US Highway 385 crosses in Burlington and after another 18 km the border between the states of Colorado and Kansas is reached.

When the numbered highways system was established in 1926, what is now Highway 24 through Colorado was designated US Route 40S . It began further west in Grand Junction and ran on today's Interstate 70. From Minturn, the further course to Limon corresponded to today's Highway 24. The further course to the eastern border of Colorado corresponded to the current route of Highway 24, but was as US Route 40N designated. In 1936, US Route 40S west of Limon and US Route 40N east of Limon merged and still form the western extension of Highway 24, which previously ended in Kansas City . The western end point of Highway 24 was moved in 1975 from Grand Junction to today's end point at Minturn.
