On June 8, 2010, the participants were divided into six groups of four teams each. The betting order was based on the results of the individual clubs in previous years. Each team played a home and an away game against the three group opponents. In the group stage, the teams received:

  • four points for a win
  • two points for a tie
  • a bonus point for four or more attempts
  • a bonus point for a loss of seven or less points

For the quarter-finals, the six group winners (classified according to the number of points won in places 1–6) and the two best runners-up (classified in places 7–8) qualified. Teams ranked 1-4 had home rights in the quarter-finals. The first played against the eighth, the second against the seventh, the third against the sixth and the fourth against the fifth. Three of the four remaining runners-up qualified for the quarter-finals of the European Challenge Cup 2010/11 .
