At the 2009 MTV Video Music Awards held at Radio City Music Hall , New York City , on September 13, 2009 , Jackson was the final highlight of a tribute show for Michael Jackson, who started the MTV in honor of the King of Pop Awards show. She sang Scream , on the screen behind her was the music video with Michael. She also paid tribute to Michael when she performed at Capital FM in December 2009 and at the Jingle Bell Ball at London's O2 Arena .

Her single Make Me was published on her website after the VMA performance and offered for download. Shortly after its release, the song became Jackson's 19th number one hit on the US dance charts, only Madonna got 40 more. Her second greatest hits compilation Number Ones was released in November 2009 by Universal Music Enterprises (UMe) and EMI Music. It debuted at number 22 on the Billboard 200, with 37,000 units sold in its first week.

After Jackson split from Island Def Jam Records, she signed a record deal with A&M Records again in 2009.

In February 2010 she took part in the charity single We Are the World 25 for Haiti , along with more than 80 other artists . She sang together with her late brother Michael: In the music video, this was inserted from the old We-Are-the-World video into the new video.

In April 2010 the film Why Did I Get Married Too? released, with Jackson in the lead role. The film grossed $ 30.1 million in its first week and debuted at number 2 on the American box office. Her role brought her a 2011 nomination in the "NAACP Image Award for Outstanding Actress in a Motion Picture" category. For the film, she recorded the song Nothing , which she released before the film was released. In May 2010, she appeared in the ninth season finale of American Idol , where she sang her hits Again , Nothing and Nasty .

Jackson performed alongside Alicia Keys and Mary J. Blige at the 2010 Essence Music Festival . In July 2010 she became the spokesperson for the fur fashion label "Blackglama", for which Lena Horne, Elizabeth Taylor , Lauren Bacall and Diana Ross had previously acted as spokespersons. According to the company, she got the job because she was "a worldwide icon of music and entertainment". In November 2010, Jackson played the lead role in the drama For Colored Girls . For her role, which received critical acclaim, she received nominations at the 2011 Black Reel Awards in the categories "Outstanding Supporting Actress" and "Outstanding Ensemble".

On November 18, 2010, Jackson announced in an interview with AOL the plan for a 2011 world tour in which she would promote and introduce her greatest hits collection Number Ones . The “Number Ones: Up Close and Personal” tour was to include concerts in 35 major cities. On February 15, 2011, she published her first book, True You: A Journey to Finding and Loving Yourself, which hit # 1 on the New York Times bestseller list the following month . In March 2011, Jackson signed a film contract with Lionsgate in order to be able to produce "independently" and "independently" films.
