Knox County was formed on January 13, 1825 from the Free Territory of Illinois and the northern part of Fulton County. It was named after Henry Knox (1750-1806), an officer in the Continental Army in the American Revolutionary War , who was later the United States' first Secretary of War under George Washington (1789-1794).
In the period from 1825 to 1838, the size of the county changed several times. The first administrative seat was until 1831 with John B. Gum in Henderson, then until 1872 Henderson, later renamed Knoxville, was the administrative seat of Knox County.

As of the 2010 census , Knox County had 52,919 people in 21,863 households. The population density was 28.5 inhabitants per square kilometer. Statistically, 2.21 people lived in each of the 21,863 households.
The racial the population was composed of 87.5 percent white, 7.2 percent African American, 0.2 percent Native American, 0.6 percent Asian and other ethnic groups; 2.5 percent were descended from two or more races. Regardless of ethnicity, 4.8 percent of the population was Hispanic or Latino of any race.
20.6 percent of the population were under 18 years old, 61.0 percent were between 18 and 64 and 18.4 percent were 65 years or older. 49.8 percent of the population was female.
The median income for a household was 39,545 USD . The per capita income was $ 20,908. 15.5 percent of the population lived below the poverty line.
