Mussy-sur-Seine is a French commune with 1,019 inhabitants (as of January 1, 2017) in the Aube department in the Grand Est region ; it belongs to the arrondissement of Troyes and the canton of Bar-sur-Seine (until 2015: canton of Mussy-sur-Seine ). The inhabitants are called Musséens .

Mussy-sur-Seine is surrounded by the neighboring communities of Plaines-Saint-Lange in the north and north-west, Essoyes in the north and north-east, Grancey-sur-Ource in the east, Charrey-sur-Seine in the south and south-east, Gomméville in the south, Molesmes in the Southwest, Les Riceys in the west and Gyé-sur-Seine in the west and northwest.
The former route nationale 71 (today's D671) runs through the municipality .
