The 64 teams were put into two pots for the draw for the first round: The amateur pot included all the teams that
- had qualified for the DFB-Pokal through the national cups (24),
- had occupied the last four places in the second Bundesliga in the 2008/09 season (4)
- had occupied the first four places in the 3rd division in the 2008/09 season (4)
Each of these teams was drawn to a team from the professional pool, which included all the teams in the First Bundesliga and the 14 highest-placed teams in the Second Bundesliga for the 2008/09 season. The teams of the amateur pot enjoy the home right. The draw for the first main round of the DFB Cup took place on June 27, 2009 in the sports show .
In the second round, an amateur and a professional pot were also drawn. Here, however, the teams that had been promoted to the Second Bundesliga were also considered professional teams. Here, too, enjoyed the amateur teams, otherwise the team first drawn into a pairing, home rights.
From the last sixteen onwards, there was only one pot to draw. Home rights enjoyed an amateur team when they played against a professional team, otherwise the team drawn first in a pairing.
