Like his older brother Siem, Luuk de Jong was born in Aigle, Switzerland, where his parents George de Jong and Loekie Raterink played volleyball . At the age of four he came to his parents' home in the Netherlands, grew up in the Achterhoek and played for the Doetinchemer Verein DZC'68 as a child . When he was ten, he moved to De Graafschap , where he went through the other youth teams and signed his first professional contract in April 2008. On November 7, 2008 he made his debut in the Eredivisie ; in the 79th minute he was substituted on for Peter Jungschläger against NAC Breda . His first match in the starting line-up was the home game against Willem II on January 17, 2009; he scored his first goal in a 2-2 draw against FC Twente on February 8, 2009, a header. Overall, he came in the season on 14 appearances and two goals in the Eredivisie.

De Jong signed a three-year contract with FC Twente in June 2009 until June 30, 2012 with an option for a further year, which was extended to 2014 in November 2010. Here he was first used on January 17, 2010; in the 90th minute he came on for Kenneth Perez . The short stint was followed by eleven more league games by the end of the season, in which he contributed two goals to the first championship of FC Twente. In the KNVB Cup 2009/10 he was successful for his team four times in five games, in the UEFA Europa League 2009/10 he scored one goal in four appearances, the goal in the 1: 4 defeat at Werder Bremen . In the 2011/12 season he scored 25 goals in 31 games for the Enscheder in the league. In total, he scored 59 goals in 119 competitive games for Twente in three years.

For the 2012/13 season , de Jong moved to the German Bundesliga at Borussia Mönchengladbach , where he signed a contract that ran until June 30, 2017. He made his debut for the Niederrhein club on August 18, 2012, when he was in the starting eleven in the 2-0 win in the first round of the DFB Cup against Alemannia Aachen . Here he gave the assist to 1-0 by Juan Arango . He made his first game in the Bundesliga on August 25, 2012 on the first day of play against TSG 1899 Hoffenheim in the starting eleven. On the 3rd matchday he then scored his first goal in the Bundesliga to mean 1: 2 against 1. FC Nürnberg . On October 4, 2012, Luuk de Jong scored the interim 1-0 lead for his team in Gladbach's Europa League group game against Fenerbahçe Istanbul , which was also synonymous with Gladbach's first goal in the European Cup in 16 years. He was subsequently absent due to a capsule injury and only celebrated his comeback on December 6, 2012, when he was substituted on in the last group game in the Europa League, again against Fenerbahce, and scored again, this time to the 3-0 final result. On January 29, 2014 de Jong was loaned to Newcastle United until the end of the season, including the purchase option .

After Newcastle United failed to use a purchase option, de Jong moved back to PSV Eindhoven for the 2014/15 season , where he received a five-year contract. He became a leading player at PSV and won the championship title in 2015 , 2016 and 2018 . In the 2019/2020 season, Luuk de Jong moved to Sevilla FC. On August 21, 2020, he and his club won the UEFA Europa League final in Cologne against Inter Milan 3-2. With his two goals in the 12th and 33rd minute, he played a key role in the success of the 6th Europa League title of FC Sevillias.
