La Mulatière is located on the western city limits of Lyon at the confluence of the Saône and Rhône . Neighboring municipalities are Lyon in the north and east, Oullins in the south and Sainte-Foy-lès-Lyon in the west.
The A7 autoroute runs through the municipality from Lyon to Marseille .

In 1885 what was then part of the commune of Sainte-Foy-lès-Lyon was raised to an independent commune.
